Biotin-PEG*-Alkyne reacts with azides via a copper-catalyzed click
reaction to produce a stable triazole linkage. The hydrophilic
polyethylene glycol (PEG) spacer arm imparts solubility in aqueous
buffers and provides a long and flexible connection that minimizes
steric hindrance involved with binding to avidin molecules. Biotin
can be subsequently used for detection or affinity
purification.
