REAGEN™ Streptomycin ELISA Test Kit is a competitive enzyme immunoassay for the quantitative analysis of chloramphenicol in feed, fish, eggs, honey, meat (beef, chicken ), milk, milk powder, serum, shrimp and urine. The unique features of the kit are:
1. Rapid (*0 minutes), and organic reagent-free extraction method.with high recovery (*0 - *5%).
2. High sensitivity (0.5 ng/g or ppb) and low detection limit (5 ng/g or ppb for honey).
3. A quick ELISA assay (less than 2 hours regardless of number of samples).
4. High reproducibility.
Procedure Overview
The method is based on a competitive colorimetric ELISA assay.
The drug of interest has been coated in the plate wells. During the analysis, sample is added along with the primary antibody specific for the target drug.
If the target is present in the sample, it will compete for the antibody, thereby preventing the antibody from binding to the drug attached to the well. The secondary antibody, tagged with a peroxidase enzyme, targets the primary antibody that is complexed to the drug coated on the plate wells. The resulting color intensity, after addition of substrate, has an inverse relationship with the target concentration in the sample.
1. Microtiter plate reader (**0 nm)
2. Vortex mixer (e.g. Gneie Vortex mixer from VWR)
3. *0, *0, **0 and ***0 mL pipettes
4. Multi-channel pipette: *****0 mL (Optional)
SAMPLE PREPARATION Be sure samples are properly stored. In general, samples should be refrigerated at **4°C for no more than **2 days. Freeze samples to a minimum of **0°C if they need to be stored for a longer period. Frozen samples can be thawed at room temps (*0 – *5°C / *8 – *7°F) or in a refrigerator before use. Preparation of 1X Sample Extraction Buffer: Mix 1 volume of *0X Sample Extraction Buffer with 9 volumes of distilled water. Honey 1. Take out 0.5 g of honey sample, add 4.5 mL of 1X Sample Extraction Buffer. 2. Put the solution in an ultrasonic bath for 5 min or mix in a shaker for *0 min. 3. Vortex for 2 min. Shake the vials upside down briefly. 4. Use *0 mL per well for the assay. Note: Dilution factor: *0. If needed, the sample can be further diluted with 1X Sample Extraction Buffer.
1. The standards contain streptomycin. Handle with particular care.
