Product Description:
This product contains Lab LastLong DNA Polymerase, dNTPs and
optimized reaction buffer at a concentration of 2×. When using,
just add template, primers, and add water to the final
concentration of Mix to 1×. This product contains red tracking dye,
and can be directly loaded for electrophoresis without adding
loading buffer; it can also be purified for subsequent operations
such as enzyme digestion, ligation, and fluorescent sequencing. The
PCR product is 3 with an A head at the end. After purification, it
can be directly cloned using LABLEAD LabpO TOPO TA series vectors
(Cat. No.: PA***1, PA***2).
Product Features:
1. Super fast amplification speed: The extension speed can reach
***0sec/kb, and PCR can be completed in as short as half an
hour.
2. Outstanding long fragment amplification ability: It can amplify
long fragments up to *0kb (including complex templates such as the
human genome).
3. Good fidelity: The fidelity is more than 3 times that of
taq.
4. Strong amplification ability: high yield, good effect for some
complex templates with special structures and templates with high
GC content.
Notes:
1. This product has a very fast amplification speed. Simple
templates such as plasmids and simple genomes can try 5s/kb
extension speed and reduce the number of cycles to further shorten
the PCR time; long fragments (greater than or equal to 3kb) or
complex templates with low yield should use ****0s/kb extension
speed or more cycles.
2. When the Lab FL yield is low, you can try to use a longer
extension time (such as *0s/kb) and a longer number of cycles (such
as *5) to increase the product yield.
3. For templates with high GC content, the pre-denaturation and
denaturation temperature of short fragments can be increased to *8
oC and/or the denaturation time can be appropriately extended. In
order to avoid DNA damage, the denaturation temperature of long
fragments should not be increased, and the pre-denaturation time
should be appropriately extended to 5min., and the denaturation
time can be extended by ***0s.
4. If the amplification template has a high GC content or the
template is complex and the amplification effect is poor, DMSO can
be added to the reaction mixture to a final concentration of 1%*8%,
and the optimal concentration can be found by increasing the
gradient by 1%. Alternatively, add betaine to a final concentration
of 1.**1.7 M. Use touchdown PCR
